Instead, the Eisernen were paired with Royale Union Saint-Gilloise, who they had already played in the group stage.
The Belgian side are not to be underestimated, however, as they are the only team to have beaten Union Berlin at their home ground, the Stadion An der Alten Försterei, this season.
"We have something to make up for in the first leg. I think it will be a very open tie between two teams with a similar style of play. Our goal must be to get a good result in the first leg," said Union Berlin's managing director Oliver Ruhnert.
However, Union Berlin are not in the best of form, having won just one of their last five competitive matches and failing to score in four of those.
The away side have also struggled in recent weeks, but ended a four-game winless streak with a 2-1 victory over Eupen at the weekend.
As usual, Union coach Urs Fischer is expected to rotate his squad, with Jerome Roussillon, Morten Thorsby and Jordan among those who could come into the starting line-up after being benched against Köln on Sunday.
