The Jamaican international gave 'Die Werkself' a brilliant start to the game with a fantastic long-range effort after four minutes, and he doubled the lead midway through the first half.
Christoph Baumgartner pulled one back for the guests in the beginning of the second half before Florian Wirtz made it 3-1 with 55 minutes on the clock.
Hoffenheim then had both Florian Grillitsch and Stefan Posch sent off before second-half substitute Lucas Alario cemented the win from the penalty spot in injury time.
The win means Leverkusen climb to the top spot, on point clear of Bayern München and RB Leipzig.
