The Black and Yellows were 1-0 up early in the second half when Hummels was adjudged to have fouled Malik Tillman in the box, and Luuk de Jong stepped up to level for the home side.
Hummels protested that he had touched the ball, but the referee was not instructed by the VAR to review the situation on the touchline screen and stood by his original decision.
After the game, Hummels vented his frustration on social media channel X, writing:
"What a joke of a penalty against us. Again! I cannot believe there can be decisions like today or against Chelsea or PSG with the VAR."
The return leg of the last-16 tie will be played at the Signal Iduna Park in Dortmund on March 13.
