The 26-year-old is sidelined with an ankle injury, which will keep him out of the rest of the league campaign.
Apart from Poulsen and Ethan Ampadu, who recently made his return from a back injury, Nagelsmann will have a full squad to pick from for Wednesday's Bundesliga encounter with Fortuna Düsseldorf.
"Basically, it looks good," Nagelsmann said at the pre-match press conference on Tuesday.
"'Yussi' will not be able to feature again this season. Ethan [Ampadu] is back in training, but he's far from being a candidate, because he has been absent for such a long time.
"Otherwise, we have the usual aches and pain that you always have after playing games, but nothing earth-shattering that, as things stand now, will restrict the involvement of any other players in tomorrow's game," he said.
RB Leipzig will be boosted by the return of Dayot Upamecano from a one-game ban, but Nagelsmann didn't want to guarantee that he will come right back into the starting eleven.
"He doesn't just come into the side automatically. I expect a reaction and a certain performance in training," he said.
