The Danish international's deal was due to run out in 2021, but it has now been extended until 2022.
"I extended my contract one year, because I think I'm a good fit for this club. We've developed, and I have developed at the same pace as the club. And I still feel like I can continue developing at this club," Poulsen said in a video posted on the club's Twitter profile.
RB Leipzig sporting director and head coach Ralf Rangnick said:
"We agreed to extend Yussuf Poulsen's contract for another year. We're very pleased! We have a strong relationship with him - everyone can see that. 'Yussi' is invaluable to us."
"'Yussi' is a prime example of what happens when talent, the right mentality, strong characteristics and good habits all come together. He has been an absolute top player for us, and we're looking forward to more goals from him," Rangnick added.
Poulsen joined 'Die Roten Bullen' in 2013 and is the club's record appearance holder, having played 209 games since then.
